凡纳滨对虾EST-SSR标记的筛选及遗传多态性检测

Screening and polymorphism of EST-microsatellite markers in Litopenaeus vannamei

  • 摘要: 为了研究我国海南地区某凡纳滨对虾(Litopenaeus vannamei)繁育群体的种质资源多样性,选取14个比利时野生对虾的EST微卫星标记,以240个中国人工选育的凡纳滨对虾个体为研究对象,检测这些标记在该养殖群体遗传多样性评估中的可行性,结果表明,有7个引物可扩增出清晰条带,其中5个引物(CNM-MG 345, CNM-MG 390, CNM-MG 437, CNM-MG 444, CNM-MG 487)具有多态性,共获得12个等位基因,平均等位基因数为2.400 0±0.547 7,平均有效等位基因数为2.060 0±0.327 8,平均观察杂合度为0.533 3±0.233 1,平均期望杂合度为0.505 2±0.082 6,平均多态信息含量为0.4090±0.0862,表明筛选出来的5个EST微卫星标记可以用于海南地区凡纳滨对虾人工繁育品种的遗传多样性分析,研究群体的多态性差异比较显著,遗传多样性处于中度水平。对我国凡纳滨对虾养殖群体和比利时凡纳滨对虾野生群体的遗传差异性进行了探究。两个群体的遗传变异可能受到地理隔离及多代定向选育的影响。研究结果为凡纳滨对虾EST微卫星遗传多样性研究、对虾人工选育及种质利用提供了可靠的科学依据。
