斜带髭鲷野生与养殖群体遗传结构的ISSR分析

The genetic structure of Nature and reared groups of Hapalogenys nitens by ISSR analysis

  • 摘要: 采用ISSR分子标记技术,对斜带髭鲷3个群体(柘林湾人工繁育群体、厦门湾人工繁育群体和厦门近海捕捞野生群体)的遗传多样性水平和群体遗传结构进行了研究.筛选出17个ISSR引物对3个群体共54个样品进行扩增,共得到145个清晰的扩增位点,其中多态性位点80个,多态位点百分率为55.17%.Popgene分析结果表明:厦门野生群体的遗传多样性水平最高(PPB=51.72%,h=0.182 6,I=0.271 9),略高于其他2个人工繁育群体的遗传多样性水平.群体间的Nei's遗传分化系数和基因流分别为:0.087 6和5.209 8.表明斜带髭鲷3个群体的遗传分化尚未达到种群的分化水平.本文通过对斜带髭鲷野生群体及人工繁育群体遗传结构和遗传多样性水平的研究,为其种质资源的科学管理提供理论依据.
